In which work by Poe do two of the main carachters have the names of Mr Hare and Mr Burke?

I think you are "Burking" up the wrong tree, if you forgive a bad pun. Neither William Burke (1792-1829) nor his nefarious partner, William Hare, are even mentioned in one of Poe's tales, let alone given the genuine role of characters. They are at least mentioned notably in Robert Louis Stevenson's 1885 tale The Body Snatcher. Perhaps that is what you seek.
