Does this combination work. Have the 420EZ, great gun, but want to go further now.

It'll work, but one of the key advantages of the 550EX flash unit - E-TTL flash - will not work. The EOS 1 supports only TTL and A-TTL flash. The 550EX supports only TTL and E-TTL flash. So an EOS 1 with a 550EX will only operate in TTL mode. This isn't a huge loss, as A-TTL isn't actually very useful, but something to be aware of.

Nonetheless you're paying a lot of money for a bunch of features you won't be able to use. The 540EZ is probably a better match for the EOS 1. Unless, of course, you plan on upgrading to a camera that can support E-TTL flash in the future. In which case a 550EX makes sense as a longer-term investment.
